上皮變間質型的轉換(Epithelial-mesenchymal transition, EMT)

本來上皮細胞之間有E-cadherin ,讓彼此的attach非常緊密,另外有β-catenin把轉錄因子抓住(若β-catenin被破壞,則轉錄因子就會進入細胞核,最後細胞就會proliferation,ex:大腸癌)
→開始上皮-間質轉換時(EMT),E-cadherin會先loss掉,即E-cadherin的表現會減少
→細胞的tight junction喪失,loss of apico-basal polarity
→gain of mesenchymal protein(ex:fibronectin);同時細胞骨架重組,變得像偽足一樣,讓cancer cell具備運動能力
→cancer cell成功穿透
 Biomarkers of EMT
1. E-cadherin(Epithelial marker)↓
2. Fibronactin, vimentin(Mesnchymal markers)↑
3. Snail1, Snail2(Slug), ZEB1, ZEB2(SIP1)(Transcription factor)↑:Snail1, Snail2(Slug), ZEB1, ZEB2(SIP1)均為轉錄因子,主要是因為轉錄因子增加,會抑制E-cadherin的表現量
※ 只有E-cadherin(Epithelial marker)↓,不叫做EMT
